Antibodies to Cytokines


Cytokines are relatively small soluble proteins that regulate immune and inflammatory responses through receptor mediated mechanisms. This class of immunomodulators includes the interleukines, colony stimulating factors, tumor necrosis factors, transforming growth factors, interferons, and chemokines. Interferons can stimulate antiviral immune responses, and recombinant pegylated alpha-interferons are now being used widely and successfully for therapy of chronic hepatitis B and C. Other cytokines such as tumor necrosis factor alpha (TNFa) and IL-1 are proinflammatory and are targeted by novel antiinflammatory therapeutic antibodies such as Remicade™ and Humira™.

When developing antibodies against cytokines, neutralizing activity (i.e. inhibition of the factor's activity) is usually desired. Therefore, monoclonal antibodies against these factors are made using standard immunization and fusion methodology, then neutralizing antibodies are selected for in an applicable functional bioassay.
