Monoclonal Antibody Discovery Process

Immunization --> To generate a polyclonal antibody response in animals
Hybridoma Library --> To immortalize the antibody response as hybridomas
Monoclonal Cultures --> To isolate monoclonal antibodies from the hybridomas

1. Immunization

Immunization of mice or rats to generate a polyclonal antibody response.

Serum is collected for analysis.

PolyExpress™ - Proprietary protocol for a quick antibody response to antigen.
PolyMax™ - Continued immunization to maximize Ab affinity and concentration - final step for most antigens.
PolyExtend™ - Extended immunization for weaker antigens to further enhance the antibody response.

Mice

Mice are the standard choice for monoclonal antibody development. Three mice per protocol.

*All PolyExpress include pre-immune serum sample.

Rats

Rats are a good choice for mouse-derived antigens and provide larger quantities of sera than mice. Three rats per protocol.

*All PolyExpress include pre-immune serum sample.

The minimum antigen concentration required is 100µg/ml.

2. Hybridoma Library^TM

Creation of hybridomas from the immunized animals.

Final boost (activation) of immune cells, fusion with myeloma cells, selection of hybridomas and cryopreservation.
Obtain an immortal, cryopreserved Hybridoma Library^TM and antibody sample of the library for analysis.

*Includes final boost, fusion, HAT selection, cryopreservation of the Hybridoma Library and antibody sample.

3. Monoclonal Antibody Cultures

Creation of monoclonal cultures and antibodies from the hybridomas.

Preparation of 910 monoclonal cultures from the Hybridoma Library^TM using flow cytometry cell sorting for single viable cells.
